printedsolid - Get netfabb. It calculates volume and has repair tools.
This should be in anyone's toolbox who is working with STLs for 3D printing.

zeusminos - You can use blender. Blender has a nice 3d printing plugin that will provide you the volume among some other useful tools

enidmorse - I loaded a 6" x 6" x 6" cube into Cura. Estimated print time (0.5mm nozzle, 50mm/s, 0.33mm layer height, 100% fill) is 76 hours. Speaking from experience, that seems about right. Maybe some printers are a bit faster but there is no way you could print that in less than 50hrs!
